Resumo No atual contexto de patógenos fúngicos resistentes emergentes tais como Candida albicans e Candida parapsilosis, a descoberta de novos agentes antifúngicos é uma questão urgente. Esta pesquisa teve como objetivo avaliar o potencial antifúngico da 2-cloro-N-fenilacetamida contra cepas clínicas de C. albicans e C. parapsilosis resistentes a fluconazol. A atividade antifúngica da substância foi avaliada in vitro através da determinação da concentração inibitória mínima (CIM), concentração fungicida mínima (CFM), ruptura e inibição da formação de biofilme, ensaios de sorbitol e ergosterol, e associação entre esta molécula e antifúngicos comuns, anfotericina B e fluconazol. O produto teste inibiu todas as cepas de C. albicans e C. parapsilosis, com uma CIM variando de 128 a 256 µg.mL-1, e uma CFM de 512-1,024 µg.mL-1. Também inibiu até 92% da formação de biofilme e causou a ruptura de até 87% de biofilme pré-formado. A 2-cloro-N-fenilacetamida não promoveu atividade antifúngica pela ligação ao ergosterol da membrana celular fúngica, tampouco danificou a parede celular. Antagonismo foi observado ao combinar esta substância com anfotericina B e fluconazol. A substância exibiu atividade antifúngica significativa ao inibir tanto as células planctônicas quanto o biofilme das cepas resistentes ao fluconazol. Sua combinação com outros antifúngicos deve ser evitada e seu mecanismo de ação deve ser estabelecido.

Abstract In the current context of emerging drug-resistant fungal pathogens such as Candida albicans and Candida parapsilosis, discovery of new antifungal agents is an urgent matter. This research aimed to evaluate the antifungal potential of 2-chloro-N-phenylacetamide against fluconazole-resistant clinical strains of C. albicans and C. parapsilosis. The antifungal activity of 2-chloro-N-phenylacetamide was evaluated in vitro by the determination of the minimum inhibitory concentration (MIC), minimum fungicidal concentration (MFC), inhibition of biofilm formation and its rupture, sorbitol and ergosterol assays, and association between this molecule and common antifungal drugs, amphotericin B and fluconazole. The test product inhibited all strains of C. albicans and C. parapsilosis, with a MIC ranging from 128 to 256 µg.mL-1, and a MFC of 512-1,024 µg.mL-1. It also inhibited up to 92% of biofilm formation and rupture of up to 87% of preformed biofilm. 2-chloro-N-phenylacetamide did not promote antifungal activity through binding to cellular membrane ergosterol nor it damages the fungal cell wall. Antagonism was observed when combining this substance with amphotericin B and fluconazole. The substance exhibited significant antifungal activity by inhibiting both planktonic cells and biofilm of fluconazole-resistant strains. Its combination with other antifungals should be avoided and its mechanism of action remains to be established.

RESUMO Neste trabalho foram desenvolvidos modelos matemáticos que permitem simular a produção de mel de uma determinada região. Esses modelos são baseados em equações diferenciais ordinárias e tiveram como ponto de partida o modelo neoclássico de Lucas 23 para o crescimento macroeconômico. Na determinação dos parâmetros dos modelos foram utilizados métodos numéricos de otimização não linear. Como resultado da solução numérica do modelo, foram simuladas a produção de mel de quatro municípios. Obteve-se, ainda, o índice de tecnologia aplicado a produção de mel de cada localidade, por um período de onze anos. Por meio desse índice é possível ordenar as municipalidades em termos de aplicação de tecnologia. Essa é uma das razões desse índice ser um indicador de vital importância para a gestão do agronegócio.

ABSTRACT In this work, mathematical models were developed which allow the simulation of the honey production at a certain municipality. These models are based in ordinary differential equations and were based on the neoclassical macroeconomic model by Lucas 23 for macroeconomic growth. In the models’ parameters determination, nonlinear numeric optimization methods were employed. As a result of the numerical solution of the model, honey production from four municipalities was simulated. It was also obtained the technology level attained by the honey production of a certain place, by a period of eleven years. Through this index it is possible to order the municipalities in terms of technology application. This is one of the reasons why this index is an indicator of vital importance for agribusiness management.

Este estudo testou a hipótese de que maior eficiência dissipativa de espécies arbóreas tropicais poderia apresentar vantagem ecológica no processo de sucessão florestal. Medidas diárias de trocas gasosas de uma espécie pioneira (Guazuma ulmifolia) e de uma espécie secundária tardia (Cariniana legalis) foram realizadas em plantas irrigadas e com deficiência hídrica. Análises da conectância da rede de trocas gasosas (Cg) e da autonomia (At) da planta em relação ao ambiente foram realizadas com o objetivo de verificar mudanças metabólicas na rede em resposta à deficiência hídrica. Como estimativa global da dissipação de calor latente, foi avaliada a capacidade de manutenção e resfriamento da temperatura foliar diante das mudanças na temperatura do ar (deltaT = TºCar - TºCfolha). As mudanças observadas nos parâmetros sistêmicos (Cg e At) e fisiológicos em resposta à perturbação ambiental, associadas à discrepante taxa de crescimento entre as espécies, sugeriram que a formação inicial do dossel de uma clareira por espécies pioneiras poderia ser resultado das maiores taxas de fotossíntese dessas espécies, e não necessariamente decorrentes do fato de as espécies secundárias tardias não suportarem o ambiente heterogêneo da clareira. Nossos resultados indicam que os maiores valores de assimilação de CO2 das espécies pioneiras são conseqüência da maior eficiência da estrutura dissipativa como um todo, envolvendo os processos de degradação e dissipação de energia, desde que não haja restrição hídrica. Como resultado prático, nosso estudo sugere a análise de deltaT para avaliar a eficiência da estrutura dissipativa e auxiliar na caracterização dos grupos funcionais.

This study tested the hypothesis that dissipative efficiency of tropical tree species could be an ecological advantage in the forest succession process. Daily leaf gas exchanges of a pioneer species (Guazuma ulmifolia) and a late successional species (Cariniana legalis) were evaluated under well-irrigated conditions and by withholding irrigation. Analyses of network connectance (Cg) and plant autonomy (At) were carried out in order to assess metabolic network changes in response to environmental perturbation. As a global estimation of latent heat dissipation, the capacity to both maintain and cool leaf temperature in response to air temperature changes (deltaT = TºCair - TºCleaf) was evaluated. The changes observed in both the systemic parameters (Cg and At) and the physiological ones brought about by water deficit, associated with discrepant growth rates between both species, suggested that the initial formation of gap canopies composed by pioneer species could simply be a result of the higher photosynthetic rates of these species, and not necessarily because late successional species cannot cope with such a heterogeneous environment as that of a gap. Our results indicate that, in the absence of water constraints, the highest CO2 assimilation rates of pioneer species are supported by the efficiency of the whole dissipative structure, involving both degradation and dissipative processes. As a practical result, our study suggests the deltaT analysis in order to evaluate the efficiency of dissipative structures and as a aid in characterizing functional groups.

In this note we use elementary facts from 3-dimensional linear algebra to demonstrate the formula for area changes under arbitrary linear maps A : R³ -> R³. We do it in general, from a hint given by the well known invertible case, which is an important result used in continuum mechanics. An intrinsic formula for the case when rank A = 2 is obtained and we finish by showing that we are really exploring a property of the cofactor matrix of A for an orthonormal basis.
